I am a PhD student investigating the molecular determinants of synaptic vesicle (SV) pool organisation. While SV clustering in the presynaptic bouton has been repeatedly observed, it is still unclear as to what regulates the association or dissociation of a vesicle from the cluster. SV fusion with the plasma membrane upon stimulation, and their subsequent recycling and return to the vesicle cluster is an integral part of neurotransmission (the release of neurotransmitters from the bouton into the synaptic cleft). My investigation of SVs and the pools that they organise themselves into (Readily releasable pool, Recycling pool and the Reserve pool) is facilitated by techniques such as sdTIM (Subdiffractional tracking of internalized molecules) and sptPALM (single-particle tracking photoactivated localization microscopy).
