Getting the message across: Signal transduction regulation by mitogen-activated protein kinase family and their interacting partners
Protein kinase networks transmit and process biochemical information, yet how their signals are shaped by the cellular environment remains incompletely understood. Our laboratory investigates the architecture of kinase signalling pathways and how their spatial, temporal and functional specificity is encoded. In this seminar, I will present two recent stories exploring crosstalk between protein kinase signalling and cellular metabolites. The first reveals an unexpected role for the primary microcephaly protein and a kinase-signal scaffold, WDR62, in coordinating protein chaperones and purine metabolism, with implications for neural progenitor behaviour and brain development. The second examines how intracellular pH, or H⁺ ion concentration, regulates JNK signalling through effects on kinase organisation and biomolecular condensation, providing a mechanism through which cellular context may shape signalling by JNK and related kinase families involved in diverse neurological functions.
